A portable instrument was designed based on bioluminescent magnetic immunoassay. The instrument consisted of photon counting unit, central processing unit, temperature control unit and power supply unit. The temperature control equipment integrated into the instrument supplied a most suitable environment for incubation at 37 ℃, which would be more convenient for field test than large-scale heating apparatus used in laboratory. In this study, an improved simplified coupling method, in which immunomagnetic microbead coupled with biotinylated rabbit-anti-Escherichia coli antibody could capture Escherichia coli directly, took one step instead of the previous method of two steps. The Escherichia coli detecting result showed a good relationship in log-log domain between the intensity of luminescence and the concentration of Escherichia coli in the range from 23.00 CFU/mL to 2.25×107 CFU/mL with a correlation coefficient of 0.9657. The sensitivity of the detecting system was 0.4787. In the reproducibility test, the coefficient of variation was 4.48%. The instrument was portable, easy to use, low-cost, integrated with temperature control equipment and fast-testing, which would provide potential application in food hygiene and safety, environmental monitoring and disease diagnosis.
