In Vitro Interactions of Bone Marrow Cells with Carbonate and Fluoride Containing Apatites

The purpose of this study was to investigate the in vitro interactions between osteoclastlike cells and synthetic apatites. Carbonate-free (HAP), carbonate substituted (CHA), and carbonate- and fluoride-substituted (CFA) apatites were prepared by precipitation method and characterized using X-ray diffraction, infrared spectroscopy, scanning electron microscopy with energy dispersive system and thermo gravimetric analyses. Cells from the bone marrow of one-day old rabbit were seeded on the apatite substrates. After three days in cell culture, all the apatite substrates showed TRAP positive reaction. The intensity of the reaction of TRAP positive cells decreased in the order: CHA>HAP>CFA. SEM observation showed that the morphology of cells on the CFA and/or HAP discs resembled the phagocytes such as monocytes, rather than the osteoclast. These results indicate that carbonate ions in the apatite lattice encourage the cell activity while the fluoride ions inhibit phagocyte cell activity.
