1 Introduction Methionine has been shown to be the most limitingamino acidfor milk protein production in lactating dairy cows (NRC,2001).Amino acids (AAs) are building blocks of proteins.In addition,methionine is an essential AA that plays fundamental roles in protein synthesis and a number of other biochemical and cellular processes (Orellana et al.,2007;Metayer et al.,2008).Many nuclear phosphorylated proteins such as Stat5 and mTOR which are important components in the chain of events that account for milk protein synthesis.However,the details of milk protein synthesis control at the transcript and translational levels are not well known.Some of the translation factors,including eEF1A (Sasikumar et al.,2012),eEF2 (White-Gilbertson et al.,2009) and EIF2 (Toerien et al.,2010),have been proved to regulate protein synthesis though mTOR pathway.In the current study,two-dimensional gel electrophoresis (2-DE)/MS-based proteomics analysis was used to identify nuclear phosphoproteins responsible for milk protein synthesis in dairy cow mammary epithelial cells (DCMECs),and functional analyses reveal eEF1B regulates milk protein synthesis.
