A new DNA electrochemical biosensor was described for detection of the PAT gene fragment. Poly-L-lysine and polyaniline nanotube was mixed in terms of proper ratio. Then, the mixture was modified onto the CPE to form composite film. DNA probes with phosphate groups were easily immobilized on the poly-L-lysine film with positive charged interface due to the strong static affinity. Cyclic voltammetry and electrochemical impedance spectroscopy (EIS) were used to characterize the ssDNA immobilization and hybridization with its complementary DNA sequence by [Fe(CN)6]3-/4- as indicator. The conditions for preparing composite film, DNA immobilization and hybridization were optimized. The PAT gene fragment from phosphinothricin acetyltransferase gene was detected by this DNA electrochemical sensor. The dynamic detection range of the sensor to the PAT gene fragment was from 1.0×10-11 to 1.0×10-6 mol/L. A detection limit of 2.9×10-12 mol/L could be estimated.
