Subchronic Toxicity Evaluation of Diphenylchyanarsine in Rats

Twenty SD rats of each sex were gavaged with either 0, 0.161, 0.323, 0.646 or 1.292 mg/kg of DC in sesame oil 6 d/wk for 13 weeks. The results indicated that after 3 weeks of exposure, salivation and even rhinorrhagia when serious intoxication occurred in partial rats of 1.292 and 0.646 mg/kg and the mean body weight of these two groups were obviously reduced during 12 to 13 weeks. During the exposure, the mean feed intakes per 100 g body weight were obviously reduced at 1.292 and 0.646 mg/kg. Analytic results of hematological indices indicated that the changes occurred in the rats exposed to 1.292, 0.646 and 0.323mg/kg of DC: significant decreases in red-cell counts, contents of haematoglobin, hematocrits and lymphocytes in hematology, a significant increase of mean concentration of haematoglobin. Analytic results of blood biochemical indices indicated the concentrations of potassium, total protein, total bilirubin, cholesterine in 1.292 and 0.646 mg/kg obviously increased too. Histopatholoic examinations revealed that hepatocyte vacuolation and inflammatory cell infiltration, lesion of mucous membrane of urnary bladder, abscission of gastric mucosa superficial lamella cells, enlarge chronic inflammatory foci of lung and degenerated change of kidney micro tubular cells, regression change of some adrenal medulla cells were presented in 1.292 mg/kg of rats. Minor injury of tubular structures in portal area of liver and hepatocytes, as well as minor shedding of stomach mucosa superficial lamella cell and slight distension of jejunum, ileum, colon could be seen in 0.646 mg/kg. A small quantity chronic inflammatory focus could been seen in lungs of 0.323mg/kg of rats. All tests in 0.161 mg/kg of rats did not show marked abnormality. We considered by comprehensive analyses that in this condition, main target organs of toxic reaction of DC on rats (ig) were liver, stomach, kidney, and it could cause and enlarge chronic inflammatory foci of lung. Estimated dose range for NOAEL appeared to be 0.161mg/kg when administered orally (ig) for 13 weeks.
