Determination of the enhancement of lead phytoextraction by Medicago sativa assisted by phytohormones and EDTA

In this study, alfalfa plants grown in hydroponics were exposed to 40mg Pb L1, ethylenediaminetetraacetic acid (EDTA) equimolar to Ph, and 1, 10 and 100μM of the phytohormones indole-3- acetic acid (IAA) and kinetin (Kn). An additional treatment containing Pb/EDTA/IAA/Kn was set. In this treatment both hormones were used at 100μM concentration. Lead uptake by alfalfa was determined by ICP/OES and XAS was used to determine the oxidation state of Pb in roots, stems, and leaves of alfalfa plants. Plants exposed to Pb/EDTA/100μM IAA/Kn acumulated about 9 500mg (Pb)·kg dry weight mass (d wt) in leaves (2275% more Pb than the treatment containing Pb/EDTA). This result demonstrated that the mixture EDTA/phytohormones enhanced the Pb uptake and translocation from the roots to the leaves high enough to make alfalfa reach the category of a Pb hyperaccumulator plant. The XAS studies demonstrated that Pb was absorbed and transported from the roots to the leaves as Pb (II).
