摘要

A bioelectrochemical method for the determination of heavy metals has been developed using oxidase enzymes and screen-printed electrodes. The activity of the oxidase enzymes has been measured following H_2O_2 production in the presence of the substrate with a Ru/graphite working electrode polarized at +700 mV vs. Ag/AgCl. Interfence signals due to the oxidation of the enzyme substrates have been minimized by the use of a cellulose acetate membrane placed onto the electrode surface. Inhibition of the activity of enzymes such as alcohol oxidase, sarcosine oxidase and glycerol-3-P oxidase by the metal ions Hg(Ⅱ), V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ) resulted in the costruction of calibration curves for the metals in the low ppm range. Total analysis time was 10 min (5 min incubation + 5 min measuring) with CVs of 10-17 %.
