A novel modified cyclodextrin derivative, water-soluble β-cyclodextrin polymer (β-CDP) was prepared from β-cyclodextrin and epichlorohydrin through a one-step condensation polymerization. The results of FT-IR and ~(13)C-NMR showed that a water-soluble β-cyclodextrin polymer remained the cavity structure of β-CD, which make it provide with the capability of forming inclusion complexes with a variety of guest molecules. The characterization of β-CDP was investigated by XRD, and DSC, indicating that β-CDP was an amorphous compound stable to the heat. The solubility of β-CDP was increased up to 86.2 g/100ml, about 46.6 times higher than the value of β-CD (1.85 g/100ml). The hemolysis test showed the hemolytic activity of β-CDP was low in vitro, which could be used as a safety excipient for pharmaceutical formation. Due to the favorable properties of β-CDP, the application of β-CDP in poorly water-soluble drug delivery systems could be possible.
