In this study, Agrobacterium strain EHA105 containing pCAMBIA1301 was used to treat a Medicago truncatula genotype, A17. The percentage of callus induction was very low because of Agrobacterium overgrowth. There was only about 21% induction for embryo calli and embryos also were few. This result confirmed that Medicago truncatula A17 is difficult to transform and regenerate via somatic embryogenesis. It is necessary to use high frequent regenerative Medicago genotypes to establish an efficient transformation system in order to study the expression and function of the phyA gene.
