We studied mechanisms of IgG transport across primary cultured rat alveolar epithelial cell monolayers. IgG transport was rapid and saturable, yielding net absorption. Lowering temperature to 4℃ from 35℃ led to ~75% decrease in IgG transport in the apical-to-basolateral (ab) direction, while IgG transport in the opposite (ba) direction completely stopped. Excess unlabeled Fc (but not Fab, F(ab')~2, or IgY) competed significantly with labeled IgG transport in either direction. Dexamethasone treatment led to reduction of absorptive IgG fluxes after 48 hr. These data suggest that IgG transport takes place via Fc-recognizing receptors expressed in alveolar epithelial cells and that IgG absorption across alveolar epithelium is regulable by glucocorticoids.
