The conservation of herbage biomass with the aid of fermentative bacteria to produce silage for animal feedstuff is widely used. During ensilage, the anaerobic microbial conversion of fermentable carbohydrates to organic acids leads to a reduction of the pH value and thus to a preservation of plant compounds. An array of lactic acid bacteria (LAB), such as Lactobacillus plantarum, Streptococcus lactis, Enterococcus feacalis, Pediococcus pentosaceus, one kind of Bacillus subtilis for fiber decomposed, one kind of Candida utilis for organic acid, are used to buildup the ensiling bacterial inoculant. Ensiling plant material with the aid of ensiling bacterial inoculant is a common agricultural practice for conserving forages independently of the time point of harvest. In order to identify the species-specific, 16S ribosomal DNA (rDNA) sequence for the LAB and Bacillus subtilis, 26S ribosomal DNA (rDNA) sequence for the Candida utilis are obtained via polymerase chain reaction (PCR). Through the analysis of the sequences, a phyligenetic tree of the 16S rDNA region of 5 kinds bacteria was plotted.
