The long-term goal of this research is to transform single-gene traits into freshwater emergent wetland monocots such as Typha (Cattail) and Juncus (Rush) to generate plants with the ability to remove and process heavy metals from contaminated sediments and waters. The availability of genes with specific remediation activities, coupled with routine tissue culture and gene transfer systems, offer the possibility of creating wetland plants with such tailored remediation capabilities. While tissue culture and Agrobacterium-mediated transformation systems have been defined for numerous monocot crop species, to date no wetland monocots have been transformed and only a few have been regenerated. The goals of this study were to define and compare parameters for plant tissue culture establishment and regeneration of the wetland plants Typha and Juncus, and to determine if cells of these plants could be genetically transformed by Agrobacterium tumefaciens, using the marker gene GUS to monitor transient gene expression.
