A reliable, long-term stable glucose sensor was developed and evaluated for continuous measurement of glucose in a rotating wall perfused vessel (RWPV) bioreactor, where baby hamster kidney cells (BHK-21) were grown. The sensor was based on a hydrogen peroxide electrode system. The sensor was employed to continuously measure the glucose concentration of cell culture media in a RWPV bioreactor for 30 days. The steady-state response time was less than 15 minutes. The output of the sensor was converted to the glucose concentration utilizing the two-point calibration method. The development sensor was able to measure glucose continuously and accurately for 16 days using a single calibration. The glucose measurement error using this sensor was +-15 mg/dl, as measured using the Beckman Glucose Analyzer, in the period of 16 days. The factors, which influence the sensor properties such as pH of the culture media, flow rate and temperature, were also examined.
