To construct the eukaryotic expression vector of human decorin and investigate the anti-tumor effect of decorin on sarcoma cells in vitro. The eukaryotic expression vector of recombination pcDNA3. 1 ( + )-DCN was constructed. Recombinant plasmid pcDNA3.1( + )-DCN and empty plasmid pcDNA3. 1 ( + ) were stably transfected into S180 cells by liposome transfection, respectively. G418 of different concentrations was used to screen the positive transformants with RT-PCR being used to determine the expression of DCN gene. The effect of DCN on the cell cycles and apoptosis of S180 cells was analyzed by the flow cytometer. Apoptosis indexes of the group of S180 cells transfected with DCN significantly increased, with the proportion of cells in G0/G1 phase being higher than that of other control groups and the proportion of cells in G2/M and S phase being lower than that of other control groups. Human DCN can induce sarcoma cells apoptosis, which is most occurred in G0/G1 of the cell cycle.
