Association between polymorphisms of Toll-like receptor 7/8 and asthma in a southern Chinese Han Population

Background: Toll-like receptors (TLRs) recognize pathogen-associated molecular patterns (PAMPs) of microbes in the environment and link human innate and adaptive immunity. Although TLRs are involved in the pathogenesis of asthma and atopy, there is no report about the association between TLR7 or TLR8 single nucleotide polymorphisms (SNPs) and asthma. Objectives: To evaluate the effects of TLR7/8 polymorphisms on asthma risk and asthma-related phenotypes in a southern Chinese Han Population. Methods: Genotyping of each SNP was performed using multiplex PCR in conjunction with tagged array single base extension technology. We conducted case-control and case-only association studies between the selected SNPs in TLR7/8 and asthma-related phenotypes. Results: The T allele of rs5935436 SNP in TLR7 was protective from developing asthma in males [adjusted odds ratio (ORs)=0.126,95% confidence interval (CIs)= 0.016-0.995]. The homozygote AA of rs3761623 and GG of rs3764880 were positively associated with lower forced expiratory volume in one second percentage of the predicted value (%FEV<,1>) and asthma severity in female asthmatics. These results were confirmed by haplotype analysis. Conclusion: TLR7/8 polymorphisms may play a considerable role in the pathogenesis of asthma in a gender-dependent manner. It could be clinically useful both for identifying patients at risk of asthma and for preventing its occurrence.
