The Theta-Composer is a new equipment developed for preparing composites of various paniculate materials. This equipment has the features of both the vessel-rotating mixers and the high speed mixers with high shear force. Therefore, the theta-composer is expected for disrupting cells effectively. In this study, yeast cells suspended in buffer solution was disintegrated by the theta-composer using glass beads with a diameter of 1.0 mm as disruption medium. The efficiency of disruption was determined by the degree of released soluble protein. The activities of two enzymes, namely alcohol dehydrogenase (ADH) as a representative of the cell wall-bounded enzymes and glucose-6-phosphate dehydrogenase (G-6-PDH) as a representative of intracellular enzymes, have also been assessed. The rotor speed of the theta-composer had the specific point to influence the degree of disruption. ADH was released faster than G-6-PDH under the specific operating conditions. The selective release of ADH was realized by varying rotor speed, beads weight, especially yeast cell weight. The results suggest that the theta-composer can well control the selective release of desired components from the disrupted yeast cell.
