Production of poly-beta-hydroxybutyric acid (PHB) and three key enzymes related to its synthesis, beta-ketothiolase (beta-KT), NADPH-dependent acetoacetyl-CoA reductase (AAR), and citrate synthase (CS) were studied in an obligatory methanotroph Methylosinus trichosporium OB3b. When cultivated in a two-stage mode, the initial growth-stage in a complete medium and the following production-stage in a nitrate-deficient medium, M. trichosporium OB3b showed a significant difference in the cellular PHB level depending on the medium Cu availability during its growth. When Cu was available, the level was more than 50 percent of the dry mass and this was accompanied by a low CS activity and a high beta-KT activity. When Cu was deficient, on the other hand, PHB accumulation was only 12 percent and enzyme activities were the opposite.
