Introduction: Laser Assisted Skin Closure (LASC) has been shown to accelerate wound healing and to reduce scarring. However, the mechanisms involved during the cutaneous wound repair after LASC remain unclear. This study aimed to evaluate the inflammation process and the wound repair fibroplasia, particularly the involvement of granulation tissue fibroblasts (myofibroblasts). Study Design/Materials and Methods: four dorsal skin incisions were performed on back of male hairless rats (n=3). For each wound, the good apposition of the edges was obtained with buried absorbable suture. Incisions were closed by 3 different techniques: 1-Transparent Adhesive Dressing (TAD) alone, 2-Continuous Intradermal Suture (CIDS) + TAD, 3-TAD + LASC using 145J/cm~2. Similarly, the laser was applied without incision (4-Laser Alone). The last group (no laser - no incision) was used as control (5-CTRL). Clinical examination, standard histological study (Haematoxylin-Eosin-Safran) and immunohistochemical study (monoclonal antibody directed against Alpha Smooth Muscle Actin) were performed at 3 and 7 days. The densities of inflammation and immunoreactivity were qualitatively appreciated through an arbitrary scale from 0 to 3 (0=undetected, 1=low density, 2=medium density, 3=dense) in dermis and hypodermis. Results: In the laser group, healing was acclerated when compared to the CIDS and TAD groups. When compared to the CIDS and TAD groups, LASC samples display less inflammation in hypodermis. An increased number of myofibroblasts is seen in dermis and hypodermis at Day 3 and Day 7.
