DNA-Based Discrimination and Population Genetics of the Coconut Scale Insect Aspidiotus Rigidus Reyne in the Philippines

The Philippines faced a devastating coconut scale insect (CSI) outbreak damaging 2.7 million coconut palms in the Calabarzon region from 2009-2014, and in Basilan and Zamboanga until recently. Initially, the damage was thought to be caused by Aspidiotus destructor Signoret which is native to the Philippines but further morphological analysis pointed A. rigidus as the real culprit. These two species are difficult to identify morphologically, but identification is possible based on ecological features. This study aims to provide molecular genetic evidence to support the use of these ecological characteristics to differentiate the two species from one another. We also aim to estimate the migration pattern of A. rigidus from the previously reported infestation in the Philippines. We extracted DNA from a total of 595 individuals of 3 species of CSI (A. rigidus, A. destructor and A. excisus), and sequenced the mitochondrial cytochrome oxidase subunit I (COI) gene region and the nucleotide elongation factor 1α (EF-1α) gene region. Sequence data from both gene regions distinguished the A. rigidus against the A. destructor, and provided new knowledge that the source of the group of A. rigidus is different in north and south of the Philippines.
