One of the most promising concepts for efficacious peroral delivery of poorly absorbable drugs is the concept of bioadhesion. Similar to natural bacterial adhesion to the gut, some plant lectins interact with particular oligosaccharides of the glycocalyx of enterocytes. The lectin wheat germ agglutinin (WGA) forms part of wheat flour to an extent of about 300 mg/kg and specifically binds to N-acetyl-D-glucosamine-residues. WGA resists proteolytic attack in the G/I-tract and exhibits not only cytoadhesive, but also cytoinvasive characteristics (1).
