Incremental herbicide metabolism by cytochrome P450s has been proposed as the basis for multiple-herbicide resistance in late watergrass (Echinochloa phyllopogon). In order to elucidate the mechanism of multiple-herbicide resistance, we isolated P450 genes from late watergrass and compared gene expression patterns between resistant (R) and susceptible (S) biotypes after herbicide treatment. Our results show that expression of CYP71C35, CYP81A12 and CYP89B16 are highly induced in the R biotype, in agreement with the previously reported patterns of P450 activity in microsomal fractions. In addition, CYP71C35 could only be amplified from the genome of an R biotype. The up-regulation patterns and higher expression levels of these genes in the R biotype suggest that these genes could be involved in the herbicide resistance mechanism observed in late watergrass.
