This method not only selectively labeled N-terminal peptide and protein, but also increased the hydrophobicity of the protein and peptide after labeling, which will facilitate the retention of hydrophilic peptide/protein on the RP-HPLC. Adding biotin to the N-terminal provides the potential for the further enrichment of the protein and peptide of interest. The recovery of the method needs improvement.
