Keratinocyte Growth Factor Upregulates Integrin Alpha5-beta1 in Epidermal Keratinocytes And Tissue Engineered Skin Equivalents

Keratinocyte growth factor is a member of the fibroblast growth factor family. It is produced by stromal cells but effects epithelial cells. Its expression is highly upregulated during wound healing. KGF promotes migration of keratinocytes on fibronectin. Since keratinocytes bind through alpha5-beta1 integrin on fibronectin we investigated whether the increase in migration is through this subunit. Using PCR we found that KGF increases a5 mRNA expression in 2D cells cultures as well as 3 D skin equivalents. We found out that KGF enhances wound healing in a 2 dimensional scratch wound assay model. Using function blocking antibody to alpha5 blocked KGF induced increase in migration. The signaling pathway(s) through which KGF mediates alpha5 up regulation is currently under investigation in our laboratory. An understanding of the molecular interactions of KGF with integrins may enable novel strategies in engineering better tissue substitutes for the treatment of burns and chronic wounds.
