Acute lung injury (ALI) is characterized by alterations in epithelial and endothelial cell function, pulmonary edema, and atelectasis. Pulmonary microvascular endothelial cells (PMVECs) are believed to play an important role in reconstructing the damaged endothelium during lung injury. Hence, there is a need to track PMVECs to understand their role in, reforming damaged endothelium. Fluorescent protein expression is a potentially powerful tool for labeling and tracking these cells. However, attempts to detect fluorescently-labeled cells in lung tissue using standard single-wavelength fluorescence microscopy approaches have been unsuccessful, due to the high autofluorescence of the pulmonary vasculature and surrounding lung tissues.
