To study the effect and mechanism of light quality on Ganoderma lucidum,and to provide a theoretical basis for facility cultivation with light irradiation strategy,this work focused on the effect of light quality on growth and endogenous indole-3-acetic acid (IAA) metabolism of G.lucidum.The growth index,endogenous levels of IAA and enzymes related to IAA metabolism and polysaccharides content of G.lucidum were investigated in different growth periods under different light.In this study,G.lucidum strain S3 was used.Mycelium were grown on a modified potato dextrose agar (PDA) medium in petri dishes and irradiated with red,yellow,green,blue light-emitting diodes (LED),and cultured in darkness as contrast.Stock strain was inoculated to cultivation media so as to allow a further growth of G lucidum.Reactions of primordia and fruit body to different light conditions which created by red,yellow,green,blue and white color filter in the greenhouse were studied,too.Results showed that,under the blue LED,mycelium maintained a steady growth,and polysaccharides content were significantly higher than that in other treatments; under the red LED,the growth rate and endogenous IAA content both showed highest in early stage; under the green LED,mycelium owned lowest growth rate and fresh weight.Enzymes related to IAA metabolism were also significantly influenced by light quality.The activity of indole acetic acid oxidase (IAAO),peroxidase (POD) and tryptophan synthetase under the blue LED showed high in early time,but decreased later,and the IAA content was consistently at low level.Mycelium cultured under yellow LED showed highest activity of both IAAO and tryptophan synthetase,and medium IAA content.Light also makes a difference for further development of G.lucidum.Blue light irradiation could significantly promote the formation of primordia and polysaccharides accumulation.While white light irradiation could increase yield of fruit body compared to other treatments.Endogenous IAA content of G.lucidum was highest at button stage but decreased with development,while emerged a picking up after spore spraying.Red and yellow light irradiation significantly improved endogenous IAA level during the growth of fruit body.The activity of IAAO,POD and tryptophan synthetase varied differently with growth under different light.In summary,blue light irradiation was the best from the viewpoint of steady growth,formation of primordia and polysaccharides accumulation; red light irradiation improved endogenous IAA level and promoted growth of mycelium in early stage of cultivation; white light increased yield of fruit body.Light quality affects growth and regulation of the level of endogenous IAA of G.lucidum.
