Difference in Associations of Polymorphic Variants of the Serotonin Transporter Gene with Resting-state EEG in Younger and Older Adults

We have previously shown that oscillatory baseline brain activity, attention tasks, and indices of orienting reaction are associated with 5-HTTLPR and STin2VNTR polymorphisms of serotonin transporter gene in age specific way. To better understand the genetic base of aging-related differences in brain function and behavior, this study aimed to investigate the combined effects of 5-HTTLPR and STin2VNTR polymorphisms on the resting-state electroencephalographic spectral power in 8 functional frequency bands under eyes closed and eyes open conditions in younger (19-35 years old, N = 217) and older (over 55 years old, N = 107) age groups and whether these EEG effects mediate properties of attention. Based on two polymorphisms of serotonin transporter gene (5-HTTLPR and STin2VNTR), three genotypes combinations were generated to reflect different levels of gene expression (low intermediate, and high). Difference in the EEG power associated with levels serotonin transporter gene expression was found only in older subjects. Under eyes open condition, carries of combination with high expression had reduced alpha 2, 3 and beta 1 activity in comparison with other combinations mainly in the posterior brain regions. Results of mediation analysis revealed that resting-state EEG properties mediated the effects of 5-HTTLPR and Stin2VNTR genotypes combinations on orienting reaction, so that high expression genotype was associated with lower estimates of alpha and beta EEG power, which in turn were translated into more pronounced orienting reaction.
