A novel cell traction force transducer based on shearrnsensitive cholesteryl ester liquid crystals (LCTFT) hasrnbeen developed. This transducer incorporated with arncustom-built cell traction force measurement and mappingrn(CTFM) software showed improved sensitivity and spatialrnresolution in sensing traction forces of humanrnkeratinocytes. The sensing output of the LCTFT wasrndisplayed as linear deformation lines in the liquid crystalrnsurface that were induced by contraction of actinrnfilaments via the focal adhesions of cells. In this context,rnthe traction forces that were exerted locally by twornclusters of focal adhesions induced strains in bi-axialrndirections in the liquid crystal surface. By using cellrnrelaxation technique, these strains were determined andrnthey were correlated by Poisson’s ratio. With therndisplacement information, traction forces of single cellsrnwere calculated based on Hooke’s theorem and the forcedeformationrnrelationships were derived. In this study, therntransducer detected a wide range of cell traction forcesrn(~10 - 140 nN) indicating the sensitivity of the system.rnThe cell traction force maps generated by our CTFMrnsoftware at a spatial resolution of ~ 5μm showed goodrnrepresentations of localised cell traction force fields andrnmight be useful for time-based study of dynamic cellsrntractions
