Genetic Diversity of Wild Daylily in Taihang Mountain Areas Based on ISSR Markers

Daylily (Hemerocallis spp.) is widely cultivated due to its showy flowers and its adaptation to a wide range of soil and climates.There are 18 species in Hemerocallis, of which 11 are native to China.Daylily is one of the extensively bred plant genera and the cultivars are highly diverse in flower colour and form.Understanding the genetic diversity of wild germplasm can help breeders further improve daylily cultivars.Twenty-one accessions of Hemerocallis spp.were collected from ten counties along the Taihang Mountain range.The plants were evaluated at the field station of the Institute of Horticulture, Shanxi Academy of Agricultural Sciences.Significant morphological differences were observed among the accessions.Seven accessions collected were triploid and all the rest were diploid.Genetic variation within the wild collection and between the wild collection and 16 commercial cultivars (ten from USA, five from New Zealand and one from China) were analysed using six Inter-Simple Sequence Repeat (ISSR) primers.A total of 120 polymorphic bands were obtained.Genetic diversity was high among the collected genotypes with 95% polymorphic loci (PPL), but was relatively low within commercial cultivars (PPL=89%).The genetic diversity among commercial cultivars within each country were low (PPL=72% for USA and 51% for New Zealand).A dendrogram of the 37 genotypes were constructed using BioNJ method.Pairwise similarity coefficients among the accessions ranged from 0.14 to 0.73.The genotypes were classified into three groups based on our results.All wild collections and one Chinese cultivar 'daylily' were grouped in one cluster.The cultivars from USA and New Zealand were distributed into two other clusters.The wild collections were divided into two subgroups.However, the subgroups do not reflect geographic locations of the wild genotypes.These results demonstrated that ISSR profiling is a powerful method for the identification of genetic variation of daylily cultivars and wild collections.However, a larger number and higher density of natural accessions should be collected and analysed, and more primers need to be employed for characterisation of wild collections at population level.
