摘要

The genetic alteration of p16, a member of cyclin-dependent kinase inhibitor family, has been intensively investigated using the approaches of molecular biology. The homozygous deletion of p16 was reported in a large number of tumor cell lines. However, when a homogenate of a primary tumor was used for gene-deletion detection, non-neoplastic tissue admixed in specimens could contaminate the tumor DNA. In situ PCR allows a specific DNA fragment amplified within the original cell, then detected in situ. Since the cells are morphologically identified, one could know if a specific gene exists in a certain population of cells by in situ PCR techniques.
