In order to better understand and define the biologicalrnrisks of cosmic radiation, it is important to identifyrnbiomarkers for exposure and for predicting individualrnsensitivity to radiation-induced biological damage. Ourrnstudy aims at identifying new biomarkers for radiationrnexposure to low and high linear energy transfer (LET)rnradiation. We will focus on the use of gene, exon and/orrncytokine expression signatures in human peripheralrnblood mononuclear cells (PBMCs) as biomarkers. Theserndata will be integrated with those from DNA doublernstrand break (DSB) repair kinetics in order to identifyrnbiomarkers of individual radiosensitivity.rnOur preliminary data indicate that gene/exon expressionrnsignatures can be used to accurately predict radiationrndoses of below 0.1 Gy (X-rays). Additionally, specificrnexon signatures may further enhance the sensitivity ofrnprediction. Also, we found that there is a very strongrnoverlap between gene expression changes in response tornhigh doses (1 Gy) of low (X-ray) and high-LET (C-ions;rn50-75 keV/μm) irradiation. However, we also identifiedrngenes that only responded to either X-rays or C-ions,rnsuggesting that LET-specific gene signatures may alsornbe identified.
