The interaction of liposomes containing linear CPSDGRG or cyclo (-CfDGR-) with integrin α_vβ_3 expressed on human umbilical endothelial cells (HUVEC) was studied. These liposomes does-dependently inhibited the adhesion of HUVEC to vitronectin, and released encapsulated materials upon interaction with HUVEC. Liposomes containing cyclo(-CfDGR-) had a higher inhibitory effect on HUVEC adhesion to vitronectin, and faster content release than liposomes containing linear CPSDGRG. The inhibitory capacity, IC_(50), for cyclo (-CfDGR-) immobilized on the liposome surface was 17 times lower than that of cyclo (-VfDGR-) as a monomer. Incubation of HUVEC with indocyanine Cy5.5-labeled liposomes containing RGD peptide sequence provided clear fluorescence images of HUVEC, suggesting that Cy5.5-labeled liposomes containing site specific ligands are useful for the visualization of specific molecular targets in deep tissues.
