Ectomycorrhizal mushroom of Tricholoma matsutake (S Ito et Imai) Sing is called to be the king of mushrooms as it is very precious and rare, and is collected without abstention and close to annihilation. The main difficulties in face of artificial domestication are of isolation, cultivation and inoculation of pure culture of Tricholoma matsutake having symbiosis relation with pine trees. In the study the isolation was conducted from different part of mushrooms and related material, and a great of cultivation was made using 8 kinds of culture media for pure culture of Tricholoma matsutake. The method of isolation was established for pure culture of Tricholoma matsutake based on recurrence and culture character combined with DNA fingerprint comparison.
