EVALUATION OF IN SITU BIODEGRADATION OF PERCHLORATE IN A CONTAMINATED SITE

Parameters affecting perchlorate biodegradation in the Las Vegas Wash area in Las Vegas, NV were investigated at field and laboratory. Two types of tests were performed in water samples from the Wash and the contaminated site, enumeration of perchlorate degrading bacteria and anaerobic microcosm testing. Microbial enumeration was performed by colony counts using standard plateing techniques. Microcosm testing was conducted in serum bottles using standard anaerobic procedures. A mineralutrient and buffer medium was supplemented to the testing bottles and lactate was used as the electron donor. The microbial count results show that perchlorate reducers are present along the Wash and in the contaminated area and numbers range from 30.x10~3 to 7.7x10~4 counts/mL. The microbial counts were higher in the zone where the Wash meets Lake Mead. No correlation was found between the concentration of perchlorate in the Wash and the number of microorganisms present. The results of the microcosm testing show that natural biodegradation of perchlorate in the Las Vegas site is limited by the lack of an electron donor, the presence of nitrate, and the salinity levels in the area.
