We found that the calcium used to crosslink the alginate gels, and not the alginate polymer, was responsible for the observed BMDC maturation and enhanced activation with LPS. It is well-known that calcium signaling is critical for immune cell function and that simply raising intracellular calcium levels without danger signals can differentiate dendritic cells into a mature phenotype (3). Thus, it is reasonable to believe that the calcium released from the alginate gels is elevating intracellular calcium levels and activating calcium-dependent signaling pathways that lead to the enhanced expression of maturation markers such as IL-1β, MHC class II and CD86.
