Specific Interactions of Glass Fibers with Extracellular Matrix Proteins

When inhaled glass fibers reach the alveolar tissue, primary interfacial phenomena would be expected to occur between these fibers and the extracellular matrix (ECM) of the alveolar cells. We have developed a microscale in vitro system to identify and characterize ECM proteins adsorbed to the surface of these fibers. Preliminary studies have focused o nthree fiber types: MMVF 10 (rapidly dissolving borosilicate), MMVF 34 (high alumina HT), and RCF 1 (approx 50:50 alumina:silica). Adherent ECM proteins stripped from the fibers represented a subset of ECM proteins. While most of the adsorbed proteins were common to all fiber types, some differnecess in protein profiles were observed. In several cases the amount of protein adsorbed to the fiber appeared significnatly enhanced over the amount of the same protein in the ECM. These data suggest that the fiber surfaces had high affinity for some ECM. These data suggest that the fiber surfaces had high affinity for some ECM proteins. Numerous adsorbed proteins were in the low molecular weight range which includes growth factors, hormones, and signal molecules involved in both tissue inflammatory and healing responses.
