Sex is a potent modifier of the cardiovascular system as males and females differ in several aspects of the biology and physiology of the heart. Pre-menopausal women possess a distinct advantage over men in the incidence of cardiovascular disease, however males, as opposed to females, are provided with some benefit once a negative cardiac event has occurred. The reasons for these differences are not completely understood and are likely due to many factors, but an important conclusion from these observations is that sex hormones appear to be important regulators of myocardial health. Of particular note, the sex hormones appear to influence the induction of the cardioprotective heat shock protein, Hsp70, by exercise. Male rats exhibit a robust increase in cardiac Hsp70 after a single bout of treadmill running while females tend not to show this effect unless subjected to multiple bouts or chronic endurance exercise training. While the inhibition in females has been linked to estrogen, the role of testosterone in this response has not been determined. Current studies were undertaken in the current body of work that examined the role of endogenous and exogenous testosterone treatment on the endurance exercise stress response in intact and testosterone deficient rats as well as intact female animals. Moreover, the roles of estrus cycle stage, the ovarian hormones and exercise stress markers/events including temperature in exercising female rats were determined on both the acute (i.e. mRNA by reverse transcription real time polymerase chain reaction) and longer period (i.e. protein by Western blotting) expression of the Hsp70 gene. Significant findings include the observation that testosterone is required by the male rat to garner the potentially cardio-protective increase in cardiac Hsp70 and that estrus cycle stage and ovarian hormones are correlated with the response in females. Moreover, exercise temperature is the best correlate with Hsp70 gene expression suggesting that exercise intensity plays an important role in the exercise stress response in both sexes. A better understanding of how sex hormones modify the exercise stress response has important implications in the prescription of exercise for males and females both young and old.; Keywords.
