Emdogain, an enamel matrix derivative (ENID), has been advocated for use in periodontal regeneration, as an alternative to conventional periodontal therapy, in an effort to try to mimic the events that take place during dental root development. In order to delineate possible intracellular signal transduction pathways involved in this process, we determined the effect of EMD on Protein Kinase C (PKC) isoform expression during the differentiation of MC3T3-E1. Results. EMD down-regulated the expression of PKC alpha and eta compared to controls. Cells in osteogenic media and EMD showed up to two fold increase in PKC alpha expression whereas there was up to two fold decrease in the expression of PKC eta at day 14 as compared to control. Stimulation with EMD up regulates Collagen (Col) gene expression in day 1 that gradually decreased through day 21. There was a twofold increase in Bone sialoprotein (BSP) expression at Day 14 and a maximum level of expression of Osteocalcin (OCN) at day 21. PKC alpha and eta are mainly located in the nucleus of the cells, upon stimulation with ENID, PKC alpha and eta was translocated from the nucleus and re-distribution throughout the cytoplasm. Conclusion. EMD has differential as well as biphasic effects on the differentiating osteoblast. The effect of ENID on osteoblastic cells is culture condition dependent. ENID also has differential effects depending on the differentiation phase of the osteoblast. (Abstract shortened by UMI.)
