Mycoheterotrophic plants are nonphotosynthetic and require the formation of a tripartite relationship with an autotrophic host and a mutualistic fungus to acquire carbon. To date, no study has inferred the population biology of any mycobiont in a tripartite system using ectomycorrhizal tissue from these symbioses. In this study, the genetic structure of Rhizopogon salebrosus, a mycobiont associated with Pterospora andromedea in the south-central Rocky Mountains, is reported. Ectomycorrhizae, containing both fungal and plant tissue, were collected from root balls of Pterospora andromedea. Mycobiont identification was accomplished using the ITS locus and sequences from Rhizopogon section Amylopogon found on GenBank. The genetic structure of the mycobiont was examined using phylogenetic analyses of two rDNA loci, ITS and RPB2, from 97 samples. Although P. andromedea has been found to associate with at least three Rhizopogon species throughout its range in North America, it was found to exclusively associate with the R. salebrosus species in the Rocky Mountains. The data support the presence of two genetic lineages within the R. salebrosus species group which exhibit a north-south distribution, possibly linked to historic conifer association. A potential zone of secondary contact between the two lineages was also identified and may reflect a zone of hybridization.
