Neutrophil-epithelial cell interactions in the lung after oxidant injury.

Neutrophils play a beneficial role after acute ozone exposure by removing injured airway epithelial cells and expediting epithelial repair. Studies have demonstrated this phenomenon in several species; however, the mechanisms by which neutrophils remove injured cells and the mediators important in these processes are unknown. I examined the role of two C fiber-derived neuropeptides, substance P and CGRP in neutrophil function and epithelial repair after acute ozone exposure in rats. To investigate the microenvironment and cellular events important in neutrophil removal of oxidant-injured airway epithelial cells, I developed an in vitro model of BEAS-2B,S.6 cells injured with the oxidant hydrogen peroxide. Using this model, I determined the phenotype of the airway epithelial cell(s) that upregulates IL-8 and thioredoxin, two chemokines important in neutrophil function. Additionally, I examined how neutrophils identify oxidant-injured airway epithelial cells. My research concludes that (1) substance P and CGRP are important for airway epithelial repair after acute ozone exposure but not in neutrophil dependent removal of injured cells, (2) TNFα/IL-1β induced IL-8 is retained in uninjured airway epithelial cells after oxidant injury, while (3) thioredoxin is upregulated in oxidant-injured and -stressed airway epithelial cells and (4) thioredoxin is important for neutrophil co-localization after oxidant injury.
