Chromatographic science and its recent advances have made it possible to examine some of the basic constituents of life. Evolvement of various modes of chromatography has contributed immensely in the study of protein, peptides, enzymes and nucleic acids in their purest state. High Performance Liquid Chromatography (HPLC) is the most widely used chromatographic technique today. Capillary electrophoresis (CE), because of its superior efficiencies and selectivity is becoming more popular and indeed has the potential to rival HPLC as an analytic tool in the near future. This dissertation work involved analytical method development of drug substances either in serum or dosage forms using High Performance Liquid Chromatography or Capillary Electrophoresis. Chapters 1--3 and chapters 7--9 describe analytical methods developed using HPLC. Chapters 4--6 describe analytical methods developed using capillary electrophoresis.;In chapters 1--3, direct serum injection methods were developed using Restricted Access Media HPLC columns for the determination of chlorzoxazone and its 6-hydroxy metabolite, phenylbutazone and its metabolite and some of the nonsteroidal antiinflammatory drugs (NSAIDs).;In chapter 4, a free solution capillary electrophoresis method was developed on fused silica capillaries to separate and quantitate codeine, caffeine, butalbital and aspirin in a mixture. In chapter 5, a capillary electrophoresis method was developed to separate and quantitate ephedrine (ED), theophylline (TP) and phenobarbital (PB) in a tablet dosage form.;In chapter 6, a micellar electrokinetic chromatography (MEKC) method was developed to analyze acetaminophen (AP), caffeine (CF) and butalbital (BB) simultaneously in a commercial tablet dosage form.;In chapter 7, methods were developed to separate dacarbazine and its two related impurities, 5-amino-imidazole-4 carboxamide (AIC) and 2-azahypoxanthine (AHX) by capillary electrophoresis and high performance liquid chromatography.;In chapter 8, an HPLC method was developed to determine racemic thalidomide in plasma on a avidin protein column using solid phase extraction.;In chapter 9, a commercially available avidin protein column was investigated for chiral separation of selected pharmaceuticals. Baseline resolution of enantiomers were successfully achieved for thalidomide, glutethimide, primaquine, aminoglutethimide, hydroxyzine, chlorthalidone and pyridoglutethimide.
