The pattern of incorporation of various extracellular matrix material precursors into extracellular matrix materials of developing lung tissues was investigated at stages corresponding to glandular, fetal saccular and postnatal saccular periods. Of the precursors investigated, labeling patterns with ;Most of these materials at each stage were N-linked, as judged by sensitivity to tunicamycin homologue B2. Treatment of fetal tissues with tunicamycin did not appear to inhibit branching morphogenesis. However, the expansion of terminal bud lumina seen with cultured glandular stage tissue was inhibited, indicating that these N-linked materials may play a directive role in the thinning of the terminal epithelium in the transition from glandular to saccular stages. The location of these materials during stages when lung tissues are functional may also indicate a role at these stages, perhaps as structural stabilizers for the respiratory epithelium. (Abstract shortened with permission of author.).;At the glandular stage, label was preferentially incorporated into cells of the terminal buds, but not over future airways. At the fetal saccular stage, label was preferentially incorporated into future respiratory regions, where it was localized primarily to the apical region of the epithelia. Labeling patterns at the postnatal saccular stage were comparable to those seen at the fetal saccular stage. Label incorporated at glandular stages was largely retained and terminal buds formed expanded saccular-like structures. At fetal saccular stages, label was also largely retained, but the lumina of saccules collapsed with culture. Label incorporated at postnatal saccular stages was rapidly turned over, and saccular lumina also collapsed. Clearly, the response of the tissues with regards to fate of the labeled materials under conditions of chase and of the tissue itself to culture conditions was different at each stage. These labeled materials were large, with apparent molecular weights in the range of 800-1,000 kDa.
