摘要

Sheep anti-mouse antibodies (SaMIg) and mouse antibodies have been purified by saturated ammonium sulfate salt precipitation and DEAE-Cellulose column chromatography. They were further characterised by Disc gel electrophoresis and Ouchterlony gel methods.;The sensitivity of the latex agglutination test with purified mouse IgG was about 0.005 to 1 ;The sheep anti-mouse IgG's were adsorbed onto the latex particles with various particle sizes and surface chemical groups. Passive and covalent adsorption methods have been achieved using various buffer systems and condensing reagents (spacers) respectively. The effect of pH on the adsorption of IgG's onto latex particles was also studied and the ideal pH, temperature, latex particle size and nature, purity of the antibody etc., for successful coating of the antibody have been defined.
