Epidermal Langerhans cells possess characteristic markers which may be demonstrated by immuno-histochemical or electron microscopic technique. Some studies require that the biochemical and ultrastructural characteristics of these cells be demonstrated simultaneously by an immuno-electron microscopic technique. In the initial application of this technique, specimens were embedded in an epoxy resin that was durable enough for electron microscopy. This resin was impermeable to immuno-histochemical staining. Methacrylates have been shown to be permeable to immuno-histochemical staining and durable enough for electron microscopy. This study was done to evaluate four methacrylates as embedding media for the immuno-electron microscopic examination of epidermal Langerhans cell and to determine whether these media enhance the technique. Epidermal sheets were embedded in commercially available methacrylates and processed for immuno-histochemical, electron microscopic and immuno-electron microscopic procedures. The results show that some methacrylates are suitable for either immuno-cytochemical or electron microscopic procedures, but that none of those tried were useful for immuno-electron microscopic examination of these cells.
