Three of six N.Y. Ceratocystis ulmi (Buism.) C. Moreau isolates examined using electron microscopy contained spiroplasma-like bodies. The fact that cultures of isolates which were treated with 30 ug/ml tetracycline hydrochloride lacked spiroplasma-like bodies, suggests that these bodies may be prokaryotic structures. Spiroplasma-like bodies were found to occur in C. ulmi isolates which appeared to be weakly to moderately virulent as determined from trunk inoculations of 10-year-old American elms (Ulmus americana L.). No relationship was found between the occurrence of spiroplasma-like bodies and growth form and mating type of C. ulmi. Cultural characteristic data suggests that tetracycline treatment of isolates which contain spiroplasma-like bodies may induce different physiological effects than treatment of isolates which do not contain them. Of seven C. ulmi isolates received from the University of Maine, three of four non-aggressive type isolates produced particles on a synthetic agar medium which resembled those produced by three N.Y. isolates known to contain spiroplasma-like bodies within their hyphae. Three aggressive type isolates from the University of Maine did not produce such particles. This suggests that non-aggressive type isolates may be more likely to contain spiroplasma-like bodies than aggressive type ones. DNA fluorescent Hoechst 33258 stain could not be successfully utilized to discriminate spiroplasma-like bodies from nuclei, mitochondria, and other fluorescent structures within hyphal cells.
