Characterizations of novel mechanisms for tube size control in Drosophila melanogaster.

Epithelial and endothelial tubes are fundamental components of various organs such as the lung, kidney and the vascular system. Proper size and shape of these tubes are crucial for the organ's function and any deviation from the correct size or shape result in diseases. The Drosophila trachea system is an excellent model system to study the mechanisms that control tube formation as it undergoes highly regulated tube size changes during development and is easily accessible. Previously, eight genes were identified as "tube expansion genes". Identification and characterization of the majority of the genes showed they are components of the insect septate junctions. Mutations in these genes cause abnormal length and diameter of the trachea and compromise the paracellular barrier function of the septate junctions.;Here I describe the work done to identify and characterize one of those eight genes termed convoluted. Flies mutated for convoluted have a severely defected trachea. Contrary to components of the septate junctions previously identified, it does not compromise the paracellular barrier. Cloning and further investigation of this gene reveals a surprising new pathway for tube size control.;In addition this work sheds some light on two known components of the septate junction complex required for proper tube size control. These proteins, Megatrachea and Sinuous, are members of the claudin family of proteins found in both mammals and flies.
