黄连素抑制肾腺癌细胞增殖及其分子机制

         

摘要

目的:观察黄连素对人肾癌细胞ACHN增殖的影响,并探讨其分子机制.方法:用MTT法检测黄连素处理后肾腺癌细胞增殖的变化,RIPA蛋白裂解液裂解肾腺癌细胞后提取总蛋白,BCA比色法测定蛋白浓度,将蛋白煮沸5 min使其变性,取等量蛋白进行Western blot分析.结果:10 μmol/L和20 μmol/L黄连素显著抑制肾腺癌细胞ACHN的增殖.Western blot结果显示,黄连素处理后的肾腺癌细胞c-Fos表达量明显下调.但Western blot结果未见明显的剪切后的caspase 3,说明黄连素并不诱导肾腺癌细胞的凋亡.结论:本研究初步说明,黄连素通过下调c-Fos的表达抑制肾腺癌细胞ACHN的增殖.%Objective: To observe the effects of berberine on the proliferation of human renal cancer cell line AC11N and the potential molecular mechanisms. Methods: The proliferation of renal carcinoma cells treated with berberine was detected by MTT assay. Renal carcinoma cells were lysed with ice-cold RIPA lysis buffer for extraction of the total protein, and the protein concentration in the cell lysates was quantitatively measured with BCA protein assay kit. Equal amount of samples, boiled for 5 min, was detected by Western blot assay. Results: 10 μmol/L and 20 μmol/L berberine significantly inhibited the proliferation of renal carcinoma cells AC11N. Western blot assay showed that the expression of c-Fos in AC11N cells was suppressed after treatment with berberine. Absence of cleaved-caspase 3 by Westem blot assay suggested that berberine failed to trigger the apoptosis of AC11N cells. Conclusion :The findings suggest that berberine inhibits the proliferation of renal carcinoma cells by suppressing c-Fos signaling.
