猪伪狂犬病毒C株的分离鉴定

         

摘要

One porcine pseudorabies virus was isolated from epidemiological investigation,which was named PRV C strain,then the specificity of the strain was studied.The special prime during the region gI and gE was designed to check the strain by PCR,it showed that objective band of Min A strain lied at about 5 000 bp,but the strain lied at about 1 500 bp gene sequencing showed that the sequencing was 1 538 bp.The gene deletion of the strain was during 989—4 468,about 3 570 bp.The deletion consisted of the main virulence(gE),the muta-tions happened at the site of 873,4 665,4 666,4 950.The homology of the strain and Bartha-K61 was during 95.0%—96.0%,so it was different from the Bartha-K61.The neutralization test was done on neutralization of the virus antibody by the 14 strains PRV isolated from some provinces in China and Min A strain.The neutraliza-tion indexes were 708—6 761,the strain could neutralize all the strains PRV,which indicated that it could be the candidate vaccine strain.%在流行病学调查中分离到的1株伪狂犬病毒,命名为猪伪狂犬病毒 C 株,并对其进行鉴定、纯化。在 gE 和 gI 基因段上设计特异性引物进行 PCR 检测,闽 A 株在5000 bp 处出现目的条带,该毒株在1500 bp 附近出现目的条带,对目的条带测序,扩增产物大小为1538 bp,在第989—4468位基因缺失,缺失片段大小为3570 bp,该缺失片段包含了伪狂犬病病毒的主要毒力基因(gE 基因),除此缺失片段外,分别在第873、4665、4666、4950位发生了碱基突变,同猪伪狂犬病病毒 Bartha K61同源性为95.0%—96.0%,与 PRV Bartha K61不是同一个毒株。用该毒株对部分省市分离到的伪狂犬毒株和标准毒株闽 A 株进行中和试验,中和指数为708—6761,该毒株可完全中和临床分离的伪狂犬病毒株,故可用作候选疫苗毒株。
