Using Ri plasmid of Agrobacterium rhizogenes (Pri15834), thecarnation transformation experiment with leafed stem segments of shoot from tissue culture as initial material was carried out. After transformation, explants were transferred to K0 medium with 500mg/L carbenicillin and cultured for 2~3 weeks, then non-geotropic hairy roots formed from infected wound sites at a frequency of 37.07%. Afterwards,the hairy roots were cut up and subcultured on K0,K1,K2 mediums with 250mg/L carbenicillin for 3 weeks. The results of subculture showed that the hairy roots on K0 medium grew indefinitely, those on K1 medium formed calli at a frequency of 51.42%, those on K2 medium formed green plantlets at a frequency of 25%, indicating expression of Ri plasmid integrated into the genome of carnation cell.%利用具有Ri质粒的发根农杆菌(Agrobacteriumrhizogenes)Pri15834对康乃馨组培苗的带叶茎节进行转化试验。转化后的外植体在K0附加500mg/L羧苄青霉素培养基上培养2~3周后,受感染的伤口部位长出无向地性的毛状根,频率为37.07%。将毛状根切碎分别在K0、K1、K2培养基上继续培养3周,毛状根在K0上表现为无限生长,在K1上主要表现为长愈伤组织,频率为51.42%,而在K2上则主要表现为长绿苗,频率为25%。这说明发根农杆菌的Ri质粒已整合到康乃馨细胞基因组中并得到表达。
