护肝调脂方对非酒精性脂肪性肝炎血清及肝组织瘦素、脂联素影响的实验研究

         

摘要

目的:研究护肝调脂方对非酒精性脂肪性肝炎(NASH)血清及肝组织瘦素、脂联素的影响。方法:102只雄性 Wistar 大鼠随机分为正常对照组(n =20)、模型组(n =22)、护肝调脂方高剂量组(n =20)、护肝调脂方中剂量组(n =20)、护肝调脂方低剂量组(n =20),正常对照组给予基础饲料,其余4组采用改良高脂饮食建立 NASH 大鼠模型,同时给予中药护肝调脂方干预。观察大鼠的一般情况,测量大鼠的体质量、肝重,测定血清 ALT、AST、TC、TG 的水平,测定血清及肝组织瘦素和脂联素的水平。结果:与正常对照组相比,模型组大鼠血清和肝组织瘦素含量显著升高,脂联素含量显著降低(P <0.01);而护肝调脂方治疗各组血清和肝组织瘦素含量显著降低,脂联素含量显著升高(P <0.01),其中中剂量组与低剂量组比较有统计学差异(P <0.05),高剂量组与中剂量组比较无统计学差异(P >0.05)。结论:瘦素和脂联素在大鼠 NASH 中发挥着重要作用,护肝调脂方对大鼠 NASH 具有明显的治疗作用,其作用机制可能与调节血清及肝组织中瘦素和脂联素的水平有关。%Objective: To research the effect of Hugan Tiaozhi decoction on serum and liver tissue leptin and adiponectin of nonalcoholic steatohepatitis.Methods:102 male rats were randomly divided into a normal control group (n =20),a model group(n =22), Hugan Tiaozhi decoction high (n =20), medium(n =20) and low (n =20) dose groups.The rats in control group were fed with basic diet ,and the rest in other four groups were set up by adopting the improved high -fat diet to copy NASH rat models,and at the same time they were given Hugan Tiaozhi decoction intervention . Observe the general condition, measure body weight and liver weight of the rats , determine ALT,AST,TC,TG with auto-matic biochemical analyzer, liver homogenate and biochemical determination of leptin and adiponectin levels .Results:The leptin levels in serum and liver tissue of the model group were significantly increased and the adiponectin level was decreased significantly(P <0.01).But the leptin levels in serum and liver tissue of Hugan Tiaozhi decoction group were significantly decreased and the adiponectin level was increased significantly (P <0.01).The medium dose group and low dose group were statistically significant (P <0.05), and the high dose group and the medium dose group had no statistical difference(P >0.05) .Conclusion: Leptin and adiponectin play an important role in rats with NASH .Hugan Tiaozhi decoction has an obviously therapeutic effect on rats with NASH .The mechanism may be related to regulating the levels of leptin and adiponectin on serum and liver tissue .
