Objective To investigate the protective effects of diazoxide on isletβcells injured by high glucose and high lip-id.Methods RINm5F islet cell line cultured in the media of high glucose and high lipid was treated with diazoxide.The prolif-eration of cells was detected by MTT assay and cell apoptosis by Annexin Ⅴ/PI staining and flow cytometry.The RINm5F cells were also stimulated by high glucose,and the insulin concentration of the cell supernatants was detected by ELISA.Results The proliferation of cells was significantly increased in diazoxide group when compared with the high glucose and lipid group.The apoptosis rate was(0.23±0.08)%,(1.23±0.18)%,and (0.73±0.09)% in control group,high glucose and lipid group,and diazoxide group,respectively.The insulin concentration of cell supernatants after stimulation with high glucose was (1.54±0.04),(1.00±0.21)and (1.41±0.13)μg/L,respectively,in the three groups.The insulin-secreting function was sig-nificantly increased in diazoxide group.Conclusion Diazoxide can protect islet cells against glucolipotoxicity-caused apoptosis and compromised islet function.%目的：探讨二氮嗪对高糖高脂诱导的胰岛β细胞损害的保护作用。方法用二氮嗪处理高糖高脂培养下的RINm5F胰岛细胞系,用 MTT法观察细胞增殖情况,用 Annexin Ⅴ/PI染色,流式细胞仪检测细胞凋亡,用葡萄糖刺激并用 ELISA法检测细胞上清液中胰岛素浓度。结果高糖高脂培养的细胞中加入二氮嗪后与普通高糖高脂组相比增殖活力明显增加,正常对照组、高糖高脂组及二氮嗪组细胞凋亡率分别为(0.23±0.08)%、(1.23±0.18)%及(0.73±0.09)%,二氮嗪组胰岛细胞凋亡率较高糖高脂组下降(P<0.05)。葡萄糖刺激后3组细胞上清液中胰岛素浓度分别为(1.54±0.04)、(1.00±0.21)及(1.41±0.13)μg/L,二氮嗪组胰岛素分泌功能明显升高(P<0.05)。结论二氮嗪对糖脂毒性造成的胰岛β细胞凋亡及胰岛功能衰退具有保护作用。
